In experiments with isolated mitochondria, state 2 - state 3 - state 4 transitions are used for calculating the P/O ratio and respiratory control ratio (RCR). Even with a good mitochondrial preparation, the RCR may appear to be low when observing on-line the peak oxygen flux at state 3. - There is a simple solution.
Why does the plot of oxygen flux apparently lag behind the time course of oxygen concentration?
Why does oxygen flux reach a low peak value (and a correspondingly low RCR is obtained) when an ADP pulse is added to isolated mitochondria?
- Lasting effect of EtOH-soluble inhibitors
Oxygen flux per mg tissue was significantly lower (<50 %) in one particular chamber, compared to parallel experiments in the other chamber(s). The effect was due to incomplete washout of inhibitors used in the titration protocol.
