Barometric pressure

Barometric pressure is measured by the O2k with an electronic pressure transducer and used to calculated the partial oxygen pressure in air satured with water vapour for air calibration of the POS.

 

Bench space for O2k

 

The Oxygraph-2k requires a minimum of lab space (0.5 m bench space) besides an external PC or laptop, with sufficient space for syringes and handling. For highest signal stability and optimum temperature control, the O2k should be placed in a position that excludes exposure to direct sunlight and avoids suddent changes of room temperature. Keep the space below the Oxygraph-2k bottom clear, to prevent any interference of ventilation beneath the main unit.

The chamber label of the O2k is by default A and B (left and right).  In Datlab, this provides the option to label the left and right chambers. Chamber labels are used in file names (appended to the date), as an index of the quantities (plots of concentration and flux for the left and right chamber) and protocol. When operating more than a single O2k, it is recommended to label the chambers of the second O2k as C and D, etc.

Data recording interval

 

The time interval [s] for data recording can be set between 0.2 s and 10.0 s, always identical for both chambers. Rigorous tests revealed that data recording intervals <1 s do not increase the information. The default of 2 s is an optimum for most applications. Do not decrease the Data recording interval unless necessary, since at low Data recording intervals the apparent noise of flux is increased and on-line information is reduced. The data recording interval is displayed (read only); it is selected in the Oxygraph Control window [F7]. A short data sampling interval increases the time resolution and noise of flux, which may be compensated for by selecting a low value of N for flux derivation.

Gain for POS

 

The gain can be changed for signal amplification in specific cases. The default is a gain of 2 V/µA; options are 1, 2, 4, or 8 V/µA amplifications. In particular, use a lower gain if sections of the experiment are performed at high oxygen above air saturation, when the maximum voltage is close to 10 V or higher (offscale). Use a higher gain at low signal output when the maximum voltage obtained during an experiment is <3 V (e.g. at low temperature).

High-resolution respirometry is based on the OROBOROS Oxygraph-2k, combining chamber design, application of oxygen-tight materials, electrochemical sensors and electronics, Peltier-temperature control and software features (DatLab) to obtain a unique level of quantitative resolution of oxygen concentration and oxygen flux, with a closed-chamber or open-chamber mode of operation (TIP2k).  Standardized two-point ►calibration of the polarographic oxygen sensor (static sensor calibration), calibration of the sensor response time (dynamic sensor calibration), and evaluation of ►instrumental background oxygen flux (systemic flux compensation) provide the experimental basis for high accuracy of quantitative results and quality control in HRR.

Instrumental background oxygen flux

 

Instrumental background oxygen flux in a respirometer is due to oxygen consumption by the POS, and oxygen diffusion into or out of the aqueous medium in the O2k-chamber.  It is measured in the range of experimental oxygen levels by a standardized instrumental background test, and is a property of the instrumental system.  Instrumental background correction eliminates errors by systemic flux compensation, automatically performed by DatLab.

PVDF

polyvinylidene fluoride

►Stopper; ►Stirrer bar.  PVDF is used generally in applications requiring the highest purity, strength, and resistance to solvents, acids, bases.  In the O2k, PVDF is used specifically due to its extremely low oxygen solubility, for minimization of oxygen backdiffusion into the O2k-chamber.

Rear limit of TIP2k

 

When the front or rear limit of the pump is reached, the motor stops automatically. This status is indicated by the blinking green light on the rear push button. After a short push on the TIP button in the other direction, the TIP is in the status ready, the green light is on without blinking.

Resume TIP programme

 

Resume will continue a TIP setup halted with ►Suspend. If the scheduled time for an injection was reached during the time the TIP suspended this injection will start immediately after pressing Resume.

Suspend TIP programme

 

Suspend a TIP will halt the progress of the current program line if no injection is taking place at the moment. The time counter in the status line will continue to show elapsed time for the current program line, but no injection will be started. A running injection will NOT be stopped by Suspend.

A TIP setup is a predefined set of instructions to be executed by the TIP, plus informations about the used chemicals. The entire actual setup is displayed in a table (program lines table) in the lower part of the Control window. Each setup consists of at least one (but possible many) program lines.
